TargetScout™ is an affinity chromatography-based technology using immobilized compounds that reveals and verifies specific cellular targets of small molecules. This assay is performed under mild lysis conditions with endogenously expressed, post-translationally modified full-length proteins. As a result, TargetScout™ enables proteome-wide selectivity and affinity analysis in the presence of cellular co-factors and complex partners. This approach is not target class-restricted; any type of bait (small molecules, peptides, proteins, or nucleic acids) can be immobilized for analysis, typically via amine functions or click chemistry. As one of our workhorse technologies for studying drug:target interactions, TargetScout™ provides valuable information to support many stages of drug development, including target deconvolution, lead optimization, and pre-clinical candidate selection.
In addition to single-concentration studies, concentration-resolved TargetScout™ can be used for affinity ranking of compounds and protein targets (EC50 and estimated Kd values).
